BeOne's $1B BRUKINSA Quarter Comes With a 2027 Investment Bill

A beat-and-raise, five late-stage solid-tumor shots and a stock near all-time highs — offset by another tacabrutideg filing delay and a slower margin glide path.
ONC · Earnings Call · 2026-11-05

A beat that was already in the price

BeOne's Q3 was clean by almost any operating measure. Product revenue rose 33% to roughly $1.9 billion, BRUKINSA reached $1.4 billion globally and crossed $1 billion in the U.S. for the first time, and adjusted operating income grew more than 79%. John Oyler set the tone: “Across every dimension of the business, BeOne is executing at the very highest level in our history.” — John Oyler, Chief Executive Officer · 2026-11-05 The company raised 2026 revenue guidance to $6.9 billion-$7.0 billion from $6.6 billion-$6.8 billion. “We now expect total revenue of $6.9 billion-$7 billion, compared with our prior range of $6.6 billion-$6.8 billion.” — Aaron Rosenberg, Chief Financial Officer · 2026-11-05 The asterisk is that the U.S. BRUKINSA milestone came with a timing assist. “U.S. BRUKINSA sales did have an approximate $50 million tailwind due to order timing, inclusive of an extra shipping day in the quarter.” — Aaron Rosenberg, Chief Financial Officer · 2026-11-05 Strip that out and the underlying run-rate is still exceptional, but less ahead of the tape. The stock had already rallied 33% in the prior 90 trading days and sits within 3.9% of its all-time high, so this was not a report that could win on the headline alone. The global tape has been re-rating the heme franchise: B cell malignancies rose 37.8% over 90 days across a basket that includes ONC, ABBV and GMAB. BeOne is riding a real wave, not creating one from scratch. The filed numbers confirm the operating leverage was not just call-deck math. In the last reported quarter, total revenue was $1.7 billion, up 30% year over year, and operating income was $325 million, up 227%. Free cash flow less stock comp reached $298 million. The commercial engine is real.

The superhighway moves from slogan to P&L

The real center of gravity on this call was the pipeline, and the keyword board reflects it. development superhighway became the company's top-ranked theme this quarter — ahead of mantle cell, accelerated approval and even non-small-cell lung cancer. That matters because BeOne is asking investors to value a repeatable R&D machine, not just BRUKINSA. Oyler made the case explicitly: “We have now shared differentiated data for five pivotal late-stage solid tumor programs. We have reached a level of productivity that I think is very rare in our industry.” — John Oyler, Chief Executive Officer · 2026-11-05 The five are GPC3 x 4-1BB, PRMT5, CEA ADC, CDK4 and B7-H4. The bispecific GPC3 program showed a 36% confirmed response rate in heavily pretreated HCC with a 79% six-month duration of response and no CRS. PRMT5 posted a 38.5% confirmed response rate in EGFR wild-type NSCLC and a 30.8% intracranial response rate. CEA ADC showed a 60% confirmed response rate in CEA-positive NSCLC. These are early but differentiated datasets, and BeOne is moving them into registrational work at a pace that is unusual for a company its size. The composition shift is the tell. On the prior Q3 call, Mark Lanasa framed the early solid-tumor portfolio around four programs: “we highlighted 4 programs, including CDK4, B7-H4, PRMT5 and FGFR2b that are showing very encouraging early data.” — Mark Lanasa, Chief Medical Officer (stated) · 2025-08-06 FGFR2b is no longer in the headline five; GPC3 and CEA ADC have replaced it. That is a real portfolio rotation, not just a new data readout. The new RevMed collaboration extends the model further: BeOne is funding and running the phase III RASonate 312 colorectal study and combining its PRMT5 inhibitor with RevMed's RAS assets. non small cell lung cancer is a crowded theme shared with other reporters this week, but GPC3 in HCC and the CEA ADC are more company-specific shots.

Two asterisks: tacabrutideg and the margin glide path

The first asterisk is tacabrutideg. The phase II study met its primary endpoint, but the FDA asked for more follow-up. “the FDA requested some additional follow-up. As a result of this, we are now planning to file for accelerated approval in mid-2027.” — Amit Agarwal, Medical/Clinical Executive · 2026-11-05 That is a second slip. In August, management was still pointing to a Q4 2026 filing: “we remain on track and if the data support this, we're looking forward to that AA submission in Q4 of this year.” — Amit Agarwal, Executive, likely Head of Clinical Development or similar · 2026-08-05 Earlier in 2026, the message was that timing had not changed at all. “I don't think there's been a change in terms of the timing and kind of how we're thinking about this.” — Amit Agarwal, Clinical Development Executive · 2026-02-26 The delay is not fatal — the accelerated approval is a smaller opportunity than the head-to-head pirtobrutinib study and the tacabrutideg-plus-sonrotoclax combination — but it weakens the fast-follower narrative and explains why accelerated approval is now a top-five company keyword. The second asterisk is the margin story. The CFO was direct that BeOne intends to spend into the pipeline.

while we expect operating income to continue to grow meaningfully, the pace of margin expansion is likely to be modest as compared to 2025 and 2026, as we balance near-term margin percentage gains with investment in future growth.

Aaron Rosenberg, Chief Financial Officer · 2026-11-05
That is a meaningful shift in tone from a company whose prior message emphasized operating-margin expansion of more than 10% in 2026. It does not mean the model is broken; it means the easy phase of margin expansion is over. financial flexibility is the phrase to hold onto — BeOne has effective net cash of $4.2 billion and a rising free-cash-flow base. But the company is choosing to reinvest, and R&D intensity of roughly 36% in the last filed quarter shows how much of the P&L is being plowed back into the pipeline. The tax item is a separate, non-operating boost. Management expects a valuation-allowance reversal in Q4 that will produce a material benefit to GAAP net income. Income tax became a top-10 company keyword this quarter for that reason. It is good news for reported EPS, but it is not cash and should not be capitalized like an operating margin gain.

What the tape is paying for

At roughly 68x revenue, BeOne is priced as a platform with a deep pipeline, not as a single-product biotech. The Q3 report supports that framing on the commercial side and gives investors five solid-tumor shots to underwrite. But it also confirms that 2027 will be an investment year: operating expenses grow at least at the recent pace, margin expansion slows, and the tacabrutideg filing moves right. The next catalysts are concrete — the ASH plenary for MANGROVE in frontline mantle cell, the GPC3 U.S. accelerated pathway, the upsized KANDELA-302 readout for CDK4, and the tacabrutideg head-to-head data against pirtobrutinib. The stock is near its high because the market believes the superhighway can produce approvals. The Q3 call showed the engine is real; the next twelve months must show it can carry the margin story too.
